1-1.グルタチオン代謝系の解明と鍵酵素

 バクテリアからヒトに至る生物細胞に最も普遍的に存在しているγ-グルタミル化合物はグルタチオンで、その解毒と抗酸化作用は良く知られています。そこで、グルタチオン代謝系の解明から取りかかりました。

 私たちがこの研究を始めたとき、バクテリアのグルタチオン代謝について分かっていたことは、哺乳類などと同様、γ-グルタミルシステイン合成酵素とグルタチオン合成酵素の2段階の酵素反応で生合成されるということでした。哺乳類ではグルタチオン代謝の鍵酵素としてγ-グルタミルトランスペプチダーゼ(以下、GGTと省略します)が知られていました。ヒトにおいては、GGTは肝臓疾患のマーカー酵素として血液検査に利用される重要な酵素です。そのため、GGTに関する研究報告は古くから数多くありました。しかし、γ-グルタミル結合を加水分解する反応ではなく、γ-グルタミル基を他のアミノ酸やペプチドに転移する反応が生理的に重要であると考えられていました。

 私たちは典型的なモデル微生物である大腸菌K-12株を材料に使ってGGTの研究を始めました。

私たちが明らかにしてきた大腸菌のグルタチオン代謝系についてまとめると、大腸菌は対数増殖期に生合成したグルタチオンを菌体外に排出します。いったん排出されたグルタチオンは、定常期になり栄養源が不足してくると再び菌体内に取り込まれます。菌体外からペリプラズムに入ってきたグルタチオンは、そこにいるGGTによってγ-グルタミル結合が切断され、遊離したCys-Glyが細胞質内に取り込まれます。Cys-Glyは、基質特異性が広いアミノペプチダーゼA, B, NとジペプチダーゼDによって切断され、Cys源として利用されます。この系、大腸菌にとって、Cysサルベージ系として機能しており、その最初のステップを触媒することが大腸菌におけるGGTの生理的役割であると考えられます。

 このことは、GGTがグルタチオナーゼとして働いて、細胞外のグルタチオンをCys源として利用していることを示した最初の例です。

 現在、γ-グルタミルシステイン合成酵素の脱感作機構とグルタチオンエキスポーターについて研究を進めています。
